GHK-Cu, NAD+ and Glutathione for Longevity
Interest in longevity wellness has moved beyond ordinary vitamins. GHK-Cu, NAD+ and glutathione now appear in skincare products, supplements, IV infusions and personalized “anti-aging” protocols.
But popularity is not proof.
The short answer is that all three compounds have legitimate biological roles, yet the strength of the human evidence varies considerably:
GHK-Cu has promising laboratory research and limited human evidence, mainly involving topical skin applications. Evidence for injectable GHK-Cu is insufficient.
NAD+ precursors can increase NAD-related metabolites in humans, but they have not been proven to slow human aging or extend lifespan.
Glutathione is a central antioxidant produced by the body. Some oral products can raise glutathione levels, but broader longevity benefits remain uncertain.
Injectable or IV treatments introduce additional safety, sterility and regulatory concerns.
These therapies should be viewed as emerging wellness interventions—not proven replacements for exercise, sleep, nutrition, preventive medicine or treatment of disease.
What is GHK-Cu?
GHK-Cu, also called copper tripeptide-1, is a complex formed when the naturally occurring peptide glycyl-L-histidyl-L-lysine binds copper.
Researchers are interested in GHK-Cu because laboratory and animal studies suggest it may influence:
Collagen and extracellular-matrix production
Tissue remodeling
Wound repair
Inflammatory signaling
Antioxidant defenses
Gene-expression pathways associated with cellular repair
These mechanisms make GHK-Cu especially interesting for skin aging and regenerative medicine. However, a plausible mechanism does not automatically translate into a meaningful clinical benefit.
Does GHK-Cu improve aging skin?
Topical GHK-Cu has more support than injectable use, but the evidence is still relatively limited.
Small cosmetic studies summarized in scientific reviews have reported improvements in measures such as skin firmness, density, fine lines and photodamage. However, many of these studies are older, small or difficult to evaluate independently.
In a randomized study involving patients who underwent CO₂ laser resurfacing, topical GHK-Cu did not produce significant objective improvements in redness, wrinkles or overall skin quality compared with the control regimen. Patient satisfaction was higher, but only 13 participants completed the study.
A new Phase 2 trial is evaluating topical GHK-Cu for acute wound healing, but results have not yet been reported.
Evidence verdict: Topical GHK-Cu is biologically plausible and may support cosmetic skin quality, but it should not be described as a proven systemic anti-aging treatment.
What is NAD+, and why is it connected to longevity?
Nicotinamide adenine dinucleotide, or NAD+, is an essential molecule involved in:
Cellular energy metabolism
Mitochondrial function
DNA repair
Stress-response signaling
Enzymes such as sirtuins and PARPs
NAD+ metabolism can change with age, illness and metabolic stress. This has led to interest in increasing NAD+ through precursors such as nicotinamide riboside, or NR, and nicotinamide mononucleotide, or NMN.
What is glutathione?
Glutathione is a tripeptide made from cysteine, glycine and glutamate. It is one of the body’s principal intracellular antioxidants and helps:
Neutralize reactive molecules
Recycle other antioxidants
Support detoxification enzymes
Maintain cellular redox balance
Protect proteins and membranes from oxidative stress
The liver produces glutathione naturally. Its status can be influenced by diet, health conditions, medications, alcohol exposure and oxidative stress.
What has the strongest evidence for healthy aging?
No peptide or infusion currently replaces the fundamentals associated with healthier aging:
Regular aerobic and resistance exercise
Adequate sleep and treatment of sleep disorders
A nutrient-dense dietary pattern
Avoiding tobacco
Moderate or no alcohol consumption
Blood-pressure, cholesterol and glucose management
Vaccination and evidence-based cancer screening
Maintaining social connection and cognitive activity
Emerging therapies can be studied alongside these foundations, but they should not distract from interventions already linked to better health outcomes.
